When using universal supports, why is the first coupling sometimes poor?


Universal support, and other non-nucleosidic supports, require an additional detritylation step since these require a longer reaction time. There are two means of achieving this. One is to carry out a detritylation before running your synthesis cycle, and the other is to incorporate an additional step into the synthesis cycle.
